چکیده

conclusions these results indicated a potential explanation for reactivation of hbv infection when patients with hepatitis receive rapamycin. results in hepg2.2.15 cells, hbv dna and hbsag increased when host cells were treated with rapamycin and the effect was reversed by autophagy inhibitor, 3-methyladenine (3-ma). materials and methods hepg2.2.15 cells were treated with rapamycin to induce autophagy. autophagosomes were observed by fluorescence microscopy and transmission electron microscopy. autophagy marker protein lc3-ⅱ/lc3-ⅰwas detected by western blotting. hbv dna and mrna were determined by real time pcr and southern blotting. hbsag was evaluated by elisa. objectives in this report, we studied the mechanism by which rapamycin enhances hbv replication and expression by inducing cellular autophagy. background some reports revealed that rapamycin could reactivate hbv infection. however, the mechanism has not been clearly explained.

Rapamycin Enhances HBV Production by Inducing Cellular Autophagy

BACKGROUND Some reports revealed that rapamycin could reactivate HBV infection. However, the mechanism has not been clearly explained. OBJECTIVES In this report, we studied the mechanism by which rapamycin enhances HBV replication and expression by inducing cellular autophagy. MATERIALS AND METHODS HepG2.2.15 cells were treated with rapamycin to induce autophagy. Autophagosomes were observe...

HBV Core Protein Enhances Cytokine Production

Hepatitis B virus (HBV) infection, a cause of hepatocellular carcinoma (HCC), remains a serious global health concern. HCC development and human hepatocarcinogenesis are associated with hepatic inflammation caused by host interferons and cytokines. This article focused on the association between the HBV core protein, which is one of the HBV-encoding proteins, and cytokine production. The HBV co...

Antitumor Effects of Rapamycin in Pancreatic Cancer Cells by Inducing Apoptosis and Autophagy

Rapamycin (Rapa), an inhibitor of mammalian target of Rapamycin (mTOR), is an immunosuppressive agent that has anti-proliferative effects on some tumors. This study aims to investigate the effects of Rapa suppressing proliferation of pancreatic carcinoma PC-2 cells in vitro and its molecular mechanism involved in antitumor activities. MTT assays showed that the inhibition of proliferation of PC...

The downregulation of ATG4B mediated by microRNA-34a/34c-5p suppresses rapamycin-induced autophagy

Objective(s): Autophagy-related 4B (ATG4B) plays an important role in the process of autophagy induction. However, the molecular events that govern the expression of ATG4B in this process are not well known. Materials and Methods: Human ATG4B 3'-UTR region (1377 nt) containing miR-34a/miR-34c-5p binding site was amplified by PCR. Luciferase assay was used to assess the activity of reporter gene...

Inducing autophagy

Autophagy is a lysosomal-mediated catabolic process, which through degradation of different cytoplasmic components aids in maintaining cellular homeostasis and survival during exposure to extra- or intracellular stresses. Ammonia is a potential toxic and stress-inducing byproduct of glutamine catabolism, which has recently been found to induce autophagy in an MTOR independent way and support ca...

Rapamycin, Autophagy, and Alzheimer's Disease.

Alzheimer's disease (AD) is a neurodegenerative disorder characterized by cognitive impairment and multiple pathological lesions. At the molecular level, AD is characterized by overt amyloid β (Aβ) production and tau hyper-phosphorylation. Hence, pharmacological agents that can attenuate Aβ accumulation and tau hyper-phosphorylation have potential promise for treatment of AD. Rapamycin, an inhi...
